“Development and application of time-resolved crystallography for 3D structural and dynamic studies of allosteric regulation in bacteria”

The project aims to investigate the resistance mechanism of Staphylococcus aureus penicillin-binding protein 2a (PBP2a) using serial femtosecond crystallography (SFX) at X-ray free electron lasers (XFELs). The research includes protein production, micro-crystallization, and structural analysis of PBP2a in complex with the antibiotic ceftaroline at both synchrotrons and XFELs. Time-resolved SFX experiments will capture reaction intermediates to understand the enzyme's allosteric regulation and resistance to β-lactam antibiotics. The study will contribute to antibiotic development by revealing key structural dynamics of PBP2a and aims to establish Spain as a leader in SFX-based structural biology.
